Function blocking antibodies against the human discoidin domain receptor DDR1

  • Monoclonal antibodies (mAbs) produced in mice
  • Six different mAbs all against the extracellular region of DDR1
  • Antigen: recombinant protein produced in HEK293 cells encompassing entire ectodomain of human DDR1
  • Five of the six mAbs mapped to three different epitopes in the DS-like domain of DDR1
  • Can be used for flow cytometry, immunoprecipitation, imunnohistochemistry, immunofluoresecence but do not work for Western blotting
  • All are function-blocking; they inhibit collagen-induced DDR1 phosphorylation
  • Inhibition of ligand-induced phosphorylation as bivalent mAbs and as monovalent Fab fragments (see figure below)


Anti-DDR1 mAbs block collagen-induced DDR1 phosphorylation as mAbs and as Fab fragments. (A), blocking with anti-DDR1 mAbs; (B), blocking with selected mAbs and Fab fragments. HEK293 cells expressing DDR1b were stimulated with 10 μg/ml collagen I in the presence or absence of the indicated anti-DDR1 mAbs or Fab fragments (at 10 μg/ml). Aliquots of cell lysates were analysed by SDS-PAGE and Western blotting. The blots were probed with anti-phosphotyrosine (anti-PY) mAb 4G10 (upper blots) and re-probed with anti-DDR1 Abs (lower blots).


DDR1 is also known as CD167a, CAK, CD167, DDR, EHGK2, MCK10, NEP, NTRK4, PTK3, PTK3A, RTK6 and TRKE.
